LOGBOOK · 2026-10-25 · GRADE A: FIELD & BIOASSAY TRIALS (2025-2026)

When spray trucks fail, skin repellents still work. Here is why.

Pyrethroid resistance is a sodium-channel mutation. DEET, picaridin and IR3535 do not use the sodium channel. Volatile spatial emanators sit in the middle.

Every dengue and malaria season produces the same neighbourhood rumour: local mosquitoes have evolved resistance to city spray trucks, so store-bought skin repellents no longer work either. The first half of that sentence is often true. Across Latin America, West Africa and Southeast Asia, wild Aedes aegypti and Anopheles gambiae populations carry high frequencies of knockdown resistance (kdr) mutations (such as V1016I and F1534C) in their voltage-gated sodium channels, alongside upregulated cytochrome P450 detoxification enzymes. The second half of the sentence is biochemically wrong. Topical skin repellents such as DEET, picaridin and IR3535 do not target the sodium channel at all, and 2025-2026 field trials confirm they retain 95 to 98 percent bite protection against heavily kdr-resistant vectors.

Two molecular locks: why kdr breaks sprays but not DEET

Insecticides sold for municipal fogging, residual wall spraying and standard bed nets are almost all pyrethroids (permethrin, deltamethrin, cypermethrin, lambda-cyhalothrin). A pyrethroid kills by wedging open the voltage-gated sodium channel (Vgsc) on the mosquito's nerve membrane so the nerve fires uncontrollably and paralyses the insect. When a mosquito population accumulates kdr point mutations on that channel protein, the pyrethroid molecule no longer binds tightly. In a 2026 laboratory comparison in Insects (Insects 2026, 10.3390/insects17040385), a pyrethroid-resistant Aedes aegypti strain exhibited a 457-fold topical resistance ratio to cypermethrin (and 435.3-fold in larvae), whereas a non-pyrethroid monoterpene active (geraniol) acting outside the sodium channel showed a topical resistance ratio of just 1.1.

Topical skin repellents registered by the EPA (DEET, picaridin, IR3535, and oil of lemon eucalyptus) are not nerve poisons. They act on odorant and gustatory receptors on the mosquito's antennae and labellum (including the odorant receptor co-receptor ORco and ionotropic receptor Ir40a), scrambling the plume of human lactic acid, carbon dioxide and skin volatiles at close range. Because kdr sodium-channel mutations do not alter antennal odorant receptors, a mosquito that flies straight through a deltamethrin street fog still turns away from a DEET-treated ankle.

That mechanism was tested directly in overnight human landing collections (21:00 to 06:00 GMT) in two rural communities in Ghana where wild Anopheles gambiae sensu lato carried high kdr-west allele frequencies (Parasites & Vectors 2025, 10.1186/s13071-025-06946-1). Applying 25% DEET lotion reduced mosquito bites by 95% and 20% IR3535 (sustained-release Staytec formulation) reduced bites by 98% compared with controls (p < 0.01), protecting collectors across a full 9-hour window.

FIELD & BIOASSAY COMPARISON · TARGET SITE VS PROTECTION IN KDR-RESISTANT VECTORS (2025-2026) 20% IR3535 (Staytec) topical skin lotion 98% bite reduction (9h, high kdr-west field, Ghana 2025) 25% DEET topical skin lotion 95% bite reduction (9h, ORco/Ir40a olfactory target, Ghana 2025) Transfluthrin spatial emanator (indoor) 93% bite reduction (8-wk semi-field; partial kdr cross-resistance risk) Transfluthrin spatial emanator (outdoor) 80% bite reduction (26% 24h mortality, Tanzania 2025) Contact pyrethroid spray (cypermethrin): 457x resistance ratio in kdr Aedes aegypti (Vgsc lock fails)
Topical skin repellents (green) target antennal odorant pathways (ORco / Ir40a) unmutated by pyrethroid kdr selection. Contact pyrethroids (amber-brown) target the mutated voltage-gated sodium channel (Vgsc).

The middle case: spatial emanators (transfluthrin and metofluthrin)

What about clip-on strips, passive emanator sheets and patio coils? Most passive spatial emanators release transfluthrin or metofluthrin: volatile pyrethroids that evaporate at room temperature without heating. In 2025, the World Health Organization added a conditional recommendation for prequalified indoor spatial emanators in malaria control.

Here the resistance nuance matters. Because transfluthrin is still a pyrethroid, it shares the sodium-channel target site and P450 metabolic pathways with contact pyrethroids, but delivers the molecule as an airborne vapour through the spiracles and antennae rather than through tarsal (foot) contact on a net. Two 2025-2026 datasets define the real-world boundary:

What a household should actually do

Three operational rules separate chemistry from rumour. First, if your municipality reports pyrethroid resistance in local Aedes aegypti, do not throw away your DEET, picaridin or IR3535 bottle: skin repellents operate on an independent receptor family and keep their full label hours. Second, treat passive transfluthrin spatial emanators as a semi-enclosed zone tool (porches, bedrooms) that reduces landings by 80 to 93 percent over eight weeks, not as an invisible wall in open wind. Third, remember that no chemical mutation helps a larva survive on dry concrete: the ten-minute weekly yard survey and gravid ovitraps bypass every resistance gene on earth.
